Cyclic membrane changes in animal cells: transformed cells permanently display a surface architecture detected in normal cells only during mitosis.

چکیده
A membrane site exposed only during mitosis in normal 3T3 cells, but exposed in all polyomavirus transformed 3T3 (Py3T3) cells, has been detected The site was identified by the specific binding of a wheat-germ agglutinin conjugated with fluorescein isothiocyanate. The receptor site has been studied previously. It binds wheat-germ agglutinin, causing the selective agglutination of Py3T3 cells, but not normal 3T3 cells. A short treatment with low concentrations of trypsin exposes cryptic sites in 3T3 cell membranes; this permits agglutination of these cells. The experiments reported here indicate that these sites normally are exposed during a brief period of the cell cycle for 3T3 cells. Specific hapten inhibitors of agglutination also inhibit the binding of the conjugated agglutinin and prevent fluorescence.

Cell Cycle-dependent Changes in the Surface Membrane as Detected with [3h]concanavalin A
A number of attempts have been made recently to study cell surface changes which occur during the cell cycle of tissue culture cells . A number of laboratories have reported that the expression of a variety of cell surface markers (e .g ., lectin receptor sites and antigens) change during the cell cycle (1-3) . This laboratory has previously demonstrated that normal mouse embryo fibroblasts bin...
متن کاملEffects of Over-Expression of LOC92912 Gene on Cell Cycle Progression
Background: We had previously identified the genes involved in squamous cell carcinoma of the head and neck using differential display and DNA microarray techniques. We also reported the first analytical study on a novel human gene called LOC92912, which was identified by differential display as a gene up-regulated in such carcinomas. LOC92912, which is a putative member of the E2 ubiquitin con...
متن کاملFunctional Surface Display of Laccase in a Phenol-Inducible Bacterial Circuit for Bioremediation Purposes
Background: Phenolic compounds, which are produced routinely by industrial and urban activities, possess dangers to live organisms and environment. Laccases are oxidoreductase enzymes with the ability of remediating a wide variety of phenolic compounds to more benign molecules. The purpose of the present research is surface display of a laccase enzyme with adhesin involved in diffuse adhesion (...
متن کاملNuclear Architecture and Epigenetics of Lineage Choice
Differentiation is an epigenetic process which is installed by changes of transcriptional programs over successive cellular divisions. A number of studies have reported the effects of biochemical modifications of chromatin (DNA and chromatin proteins) on the regulation of transcription. Although, these studies are able to explain how transcription of a given gene is regulated (toward activation...
متن کاملFacts about Stem Cells and Importance of Them
Stem cells are undifferentiated biological cells that can differentiate into specialized cells and can divide (through mitosis) to produce more stem cells. They are found in multicellular organisms. In mammals, there are two broad types of stem cells: embryonic stem cells, which are isolated from the inner cell mass of blastocysts, and adult stem cells, which are found in various tissues. In ad...
